本片所有場景幾乎都發生在一個小房間內，講述11名陪審團如何在另一名持反對意見的陪審團的說服下放棄成見，以挽救一條性命。 故事背景設定在1950年的美國紐約，一名在平民窟長大的西班牙裔美國少年，被控以彈弓刀殺死自己的親生父親，案發時目擊者和證物鐵證如山，少年將被處以一級謀殺的死刑。擔任此案的12名陪審團結案前在休息室商議案情，皆對此案不削一顧，認定少年一定是殺人兇手，只有8號陪審員，針對案情疑點逐一申論擊破，最後獲得其他陪審員的認同，進而扭轉裁判。
約翰歐德曼教授突然提出離職，因此眾人為他辦了一個歡送派對，在派對上約翰卻向大家坦承自己其實已經活了14,000年，而他每十年就必須換個地方生活，否則他不曾改變的面容以及他的兒女將會發現這個事實...。
